Detection of Hepatitis B Virus(HBV) DNA by Polymerase Chain Reaction(PCR)


Abstract
We have performed a rapid, simple, and economic procedure for detection of hepatitis B virus(HBV) DNA from serum and formalin-fixed, paraffin-embedded liver needle biopsy tissue using nested polymerase chain reaction(PCR) technique. HBV DNA was
released
from virions by incubating serum and deparaffinized needle biopsy tissue with alkaline solution (NaOH), and amplified with rapid hot air thermocycler. The amplified HBV DNA sequences were detected by ethidium bromide stained agarose gel
electrophoresis
after nested PCR. All procedure could be completed in 3-4 hours. We detected HBV DNA from 30(94%) of 32 HBsAg positive patient serum and 4(31%) of 13 HBsAg negative patient serum, 43(98%) of 44 formalin-fixed, paraffin-embedded liver needle
biopsy
tissues with HBsAg positive chronic active and persistent hepatitis and 7(88%) of 8 HBsAg positive alcoholic hepatitis, hepatic cell necrosis and nonspecific finding tissues. Thus this rapid nested PCR method would be useful for detecting DNA
sequences
of HBV and other viruses in serum and biospy tissues for routine laboratory diagnostic test.
